慢性高血糖是糖尿病的一个主要特征,它也是发生糖尿病并发症的一个主要病因。目前普遍认为,高血糖毒性通过影响β细胞的分泌,使得2型糖尿病更加恶化。对于慢性高血糖有害影响的机制有很多不同的解释。其中一种学说认为,在高血糖状态下,几个关键蛋白的调节可能可以解释高血糖的慢性毒性作用。因此,在研究蛋白表达对于慢性高血糖毒性的影响方面,生物标本的蛋白质组学分析是一个有趣的方法。新发现的在慢性高血糖调节后表达的蛋白质,可能对更好的研究高血糖毒性条件下的机制有帮助。在这里,将对已应用于胰腺、胰岛细胞和β细胞相关研究的蛋白质组学技术作一综述。
Chronic hyperglycemia is a major feature of diabetes and it is also a major cause of diabetic complications. It is now generally accepted that hyperglycemic toxicity worsens type 2 diabetes by affecting the secretion of beta cells. There are many different explanations for the mechanism of the harmful effects of chronic hyperglycemia. One of the theories suggests that the regulation of several key proteins may explain the chronic toxic effects of hyperglycemia in hyperglycemic conditions. Therefore, proteomic analysis of biological samples is an interesting approach in studying the effects of protein expression on chronic hyperglycemic toxicity. Newly discovered proteins expressed after chronic hyperglycemic regulation may be helpful in better understanding the mechanisms under hyperglycemic toxicity. Here, review of proteomics techniques that have been applied to pancreatic, islet and beta cell related research.
