目的本研究对我们前期利用功能蛋白质组学技术发现的吲哚美辛(Indomethacin,IN)抗结肠癌的非COX-2途径相关蛋白Bfl-1、WISP-1和PCNA的表达差异进行验证,并探讨IN抑制肿瘤的相关机制。方法应用荧光定量PCR技术检测IN处理组和未处理组HCT116细胞Bfl-1、WISP-1和PCNAmRNA的表达。结果吲哚美辛下调Bfl-1、WISP-1和PCNAmRNA的表达。结论吲哚美辛通过Bfl-1、WISP-1和PCNA等非COX-2依赖途径诱导HCT116细胞的凋亡并抑制其增殖
Objective This study validated the differential expression of non-COX-2 pathway-related proteins Bfl-1, WISP-1 and PCNA in indomethacin (IN) anti-colon cancer cells detected by functional proteomics To investigate the mechanism of IN inhibiting tumor. Methods Fluorescent quantitative PCR was used to detect the expression of Bfl-1, WISP-1 and PCNA in INT-treated and untreated HCT116 cells. Results Indomethacin down-regulated the expression of Bfl-1, WISP-1 and PCNA mRNA. Conclusion Indomethacin induces apoptosis and inhibits proliferation of HCT116 cells through non-COX-2-dependent pathways such as Bfl-1, WISP-1 and PCNA
