1.用71型分光光度计在可見光部分测得氧合血兰旦白的吸收光譜在440mμ光密度最高,随着波长的增长而下降,至570mμ达一最低值,然后逐渐回升,在570mμ达另一高峰,然后又逐漸降到510mμ的最低值以下。还原血兰旦白的吸收光譜則随着波长的增长而不断下降。血兰旦白色素团的吸收光譜在全段波长中以510mμ为最低,570mμ为最高峰。 2.用pH7.3硼砂緩冲液稀释血淋巴可以得到比較正常的血兰旦白吸收光譜曲线。水、生理盐水和0.0067N氢氧化氨不适于作稀释剂。 3.血兰旦白去纤維并稀释后用橡皮球通过滴管打入空气,每打气5—10次在570mμ試测光密度一次,直到光密度不再上升或开始下降为止,然后倒入比色管中测量其吸收光譜。这样可以在短时間內得到氧合血兰旦白溶液,并且可以避免因通气过度而引起光密度的降低,因而获得比較显著与正常的吸收光譜曲线。在稀释的血兰旦白溶液中通入氢20分钟可以得到还原血兰旦白。通氢后在液面加一层石蜡油。
