目的:探讨民族药头花蓼居群间的DNA序列变异与地理分布的关系,为头花蓼优良种质资源的筛选评价提供分子依据。方法:采用PCR产物直接测序,对头花蓼11个居群11个个体样品的叶绿体psbA-trnH,trnL-trnF基因进行测序分析研究。结果:头花蓼11个居群的叶绿体psbA-trnH长402 bp,存在6个变异位点。trnL-trnF长875 bp,存在5个变异位点。用UPGMA法构建的聚类图表明,头花蓼在贵州至云南的类群存在较大程度变异。结论:贵州西部与云南东部地区的头花蓼有利于优良种质资源的筛选。
OBJECTIVE: To investigate the relationship between DNA sequence variation and geographic distribution of Polygonum multiflorum populations and to provide a molecular basis for the screening evaluation of Polygonum capitatum germplasm resources. Methods: The chloroplast psbA-trnH and trnL-trnF genes of 11 individuals of 11 populations of Polygonum capitatum were sequenced and analyzed by direct sequencing of PCR products. Results: The chloroplast psbA-trnH in 11 populations of Polygonum capitatum was 402 bp in length, with 6 variation sites. trnL-trnF is 875 bp in length, with five mutated sites. The clustering map constructed by UPGMA method shows that there is a large degree of variation in the taxa of Polygonum capitatum from Guizhou to Yunnan. Conclusion: Polygonum capitatum in western Guizhou and eastern Yunnan is good for selection of excellent germplasm resources.
