目的:探讨不同采收月份明党参根中L-天门冬酰胺的动态累积规律。方法:采用邻苯二甲醛(OPA)柱前衍生HPLC测定明党参中L-天门冬酰胺的含量。取供试品溶液1 mL,OPA衍生试剂1 mL,硼酸缓冲液(0.2 mol.L-1,pH 10.5)5 mL,定容至10 mL,反应时间20 min;流动相:A为1.35%醋酸钠缓冲液(pH 7.3),B为1.35%醋酸钠(pH 7.2)-乙腈-甲醇(100∶175∶225)。洗脱梯度(0~20 min,7%B);20~25 min,100%B,维持8 min),柱温40℃,检测波长333 nm。结果:在质量浓度100~1 000 mg·L-1对不同采收期的样品在实验条件下测定,L-天门冬酰胺平均含量为0.26%~1.87%。结论:L-天门冬酰胺含量在5~8月及1月较高,9月至次年4月较低。
OBJECTIVE: To investigate the dynamic accumulation of L-asparagine in root of Cynanchum paniculatum collected from different months. Methods: The content of L-asparagine in Codonopsis Pilosula was determined by pre-column derivatization HPLC with OPA. Results: The average concentration of L-asparagine was between 0.26% and 1.87% at the concentration of 100-1000 mg · L-1 for different harvest periods under the experimental conditions. Conclusion: The content of L-asparagine is higher in May-August and January and lower in September-April.
