目的:探讨人巨细胞病毒重组抗原在ELISA中的应用.方法:采用基因工程方法组建含有巨细胞病毒PPUL32及PPUL44蛋白共三个强抗原决定簇基因的重组抗原克隆,以此重组抗原建立ELISA方法,用于117份待测血清巨细胞病毒IGM抗体的检测,并将ELISA方法与免疫转印方法进行了比较.结果:经统计学分析,建立的ELISA方法与免疫转印检测方法的检测结果具有非常显著的一致性(χ2=54.08,P<0.01).与免疫转印方法比较,其敏感性和特异性分别为100%和81.3%;阳性预测率和阴性预测率分别为60.5%和100%,此ELISA方法具有良好的重复性.结论:此重组抗原具有良好的应用价值,可望替代HCMV感染ELISA诊断中的全病毒抗原.“,”Objective: Our aim was to evaluate the application of human cytomegalovirus(HCMV) recombinant antigen in enzymelinked immunosorbent assay(ELISA).
