目的和方法:Heymann肾炎(HN)是一种用于研究人类膜性肾病的大鼠动物模型。用主动型HN模型大鼠的淋巴细胞与小鼠SP2/0骨髓瘤细胞融合,制备针对HN致病原的单克隆抗体c3~6(McAbc3~6),并用它诱导大鼠被动型HN。结果:免疫酶组织化学及免疫电镜显示,McAbc3~6特异性识别肾小球上皮足突细胞膜抗原并与其结合形成免疫复合物。
PURPOSE AND METHOD: Heymann nephritis (HN) is a rat animal model used to study human membranous nephropathy. Monoclonal antibody c3 ~ 6 (McAbc3 ~ 6) against HN pathogen was prepared by fusion of mouse lymphocytes from mouse model of active HN with mouse SP2 / 0 myeloma cells and used to induce rat passive HN. Results: Immunohistochemistry and immunoelectron microscopy showed that McAb 3 ~ 6 specifically recognized the membrane antigen of glomerular epithelial cells and formed an immune complex.
