目的对野生黄连进行遗传多样性研究。方法通过ISSR技术对7个野生黄连居群共78个个体进行遗传多样性分析。结果用12个随机引物共扩增出106条清晰条带,其中72条具多态性,平均多态性位点比率为67.92%,Nei′s基因多样性指数H=0.1803,Shannon多样性指数I=0.2832,遗传分化指数Gst=0.6815,遗传距离和遗传一致度分别为:0.0894~0.1846和0.8321~0.9127。结论黄连种水平上具有较高的遗传多样性,遗传变异主要存在于居群间,遗传多样性与地理关系表现出明显的相关性,ISSR可以作为研究遗传多样性及遗传分化的有效标记。
Objective To study the genetic diversity of Coptis chinensis. Methods The genetic diversity of 78 individuals of seven populations of Coptis chinensis was analyzed by ISSR. Results A total of 106 clear bands were amplified with 12 random primers. Among them, 72 were polymorphic with an average of 67.92% loci, Nei’s gene diversity index H = 0.1803, Shannon’s diversity index I = 0.2832, Gst = 0.6815, genetic distance and genetic identity were 0.0894 ~ 0.1846 and 0.8321 ~ 0.9127, respectively. Conclusion The genetic diversity of Coptis Chinensis has high genetic diversity. The genetic variation mainly exists in the populations. The genetic diversity and geographical relationship show obvious correlation. ISSR can be used as an effective marker for studying genetic diversity and genetic differentiation.
