对血浆、组织及生物体液中游离脂酸(FFA)的测定,目前用得多的为比色法。此法依据的基本原理是:游离脂酸可与铜或钴形成脂肪酸的铜盐或钴盐,与一定的显色剂作用而产生有色化合物,颜色的深浅与脂酸的含量成正比。脂酸的铜盐(或钴盐)是被抽提在有机溶剂相中,由于不同测定方法设计上的差异,操作过程中此有机相可以是上相,亦可能是下相:前者不仅操作方便,且不易被过量的铜或钴所污染以致测定结果偏高;后者弊病多端,为此我们对FFA的测定方法作了改良。 一、材料和方法 1.本法作如下改良:①脂酸提取:以石油醚代替Dole氏溶液中的庚烷,抽提液分相快,脂酸被提取在上相,去磷脂及有机酸的效果较好。②显
Determination of free fatty acids (FFA) in plasma, tissues, and biological fluids is currently much more colorimetric.
