分离群体是甘蔗基因发掘的重要前提条件,开展甘蔗分离群体数量性状及分子标记数据的评价研究,为甘蔗遗传连锁图谱构建和数量性状基因定位及遗传育种提供依据。对甘蔗杂合商业品种与野生种割手密远缘杂交的F1群体(Co419×Y75/1/2)的产量性状(有效茎数、单茎重、株高、茎径、公顷产量)和糖分性状(蔗汁糖度、锤度、商业糖分)的频率分布、变异系数进行分析评价;同时对12对AFLP引物和36对SSR引物获得的505个分离标记进行χ2检测。研究结果表明,F1群体的产量和糖分性状符合正态分布,变异丰富,并表现为明显的超亲分离,属于多基因控制的数量性状。AFLP和SSR标记的分离比例检测表明,群体单双剂量标记的检出比例为52%,共获得179个单剂量标记和82个双剂量标记,其中有24对AFLP和SSR引物的单双剂量标记检出比例大于50%,属于比较有效的PCR引物。通过本研究,确认了该群体适合开展遗传连锁作图和数量性状基因定位研究;同时单双剂量标记的获得和高效引物的筛选都将为后续遗传学研究奠定基础条件。
Separation of populations is an important prerequisite for sugarcane gene discovery. The quantitative characterization and molecular marker data analysis of sugarcane population were carried out to provide basis for genetic linkage map construction and quantitative trait gene mapping and genetic breeding of sugarcane. The yield traits (effective stalks, single stem weight, plant height, stem diameter, yield per hectare) and sugar content of F1 population (Co419 × Y75 / 1/2) crossed with commercial varieties of wild and sugarcane, The frequency distribution and coefficient of variation of characters (sugar juice, brix, commercial sugar) of cane juice were analyzed and evaluated. Two hundred and fifty five markers obtained from 12 pairs of AFLP primers and 36 pairs of SSR primers were used to detect χ2. The results showed that the F1 population’s yield and sugar traits were in a normal distribution, with abundant variation and obvious super-pro-segregation, belonging to quantitative traits controlled by multiple genes. The detection of AFLP and SSR markers showed that the detection rate of population single-and double-dose markers was 52%, a total of 179 single-dose markers and 82 double-dose markers were obtained, of which 24 pairs of AFLP and SSR primers single- Detection rate is greater than 50%, is a more effective PCR primers. Through this study, it was confirmed that the population was suitable for genetic linkage map and quantitative trait loci. Meanwhile, both single-and double-dose labeling and efficient screening of primers would lay the foundation for subsequent genetic studies.
