目的:比较龟鹿二仙胶及其拆方含药血清对硝普钠(SNP)诱导后软骨细胞bax、bcl-2表达的影响,探讨龟鹿二仙胶的配伍机理。方法:取第三代4周龄SD大鼠关节软骨细胞,经SNP诱导凋亡后,采用免疫组化染色法及Realtime-PCR观察龟鹿二仙胶及其拆方含药血清对软骨细胞bax、bcl-2表达的影响。结果:龟鹿二仙胶及其拆方各组均可引起bcl-2表达升高及bax下降,与生理盐水组比较差异有统计学意义(P<0.05),龟鹿二仙胶与龟板、鹿角比较差异无统计学意义(P>0.05)。结论:龟板、鹿角、人参、枸杞均具有抑制软骨细胞凋亡的作用,而4味药同用,人参、枸杞并无显著加强龟板、鹿角的抑制凋亡作用。
OBJECTIVE: To compare the effects of Guilu Erxian and its disassembled prescription serum on the expression of bax and bcl-2 in chondrocytes induced by sodium nitroprusside (SNP), and to explore the compatibility mechanism of the two compounds. Methods: The third generation of 4-week-old SD rat articular chondrocytes were induced by SNP. After induced by SNP, immunohistochemical staining and Realtime-PCR were used to observe the effects of Gulu Erxian Ginseng and its demolition prescription serum on bax , Bcl-2 expression. Results: Guilu Erxian gum and its disassembled groups could induce the increase of bcl-2 expression and the decrease of bax, compared with the normal saline group (P <0.05), Guilu Erxian gum and turtle shell, Antlers no significant difference (P> 0.05). Conclusion: The turtle shell, antler, ginseng and wolfberry all have the effect of inhibiting chondrocyte apoptosis. However, the combination of 4 herbs and ginseng and wolfberry did not significantly enhance the inhibition of the apoptosis of turtle shell and antler.
