紫外分光光度法测定温肺化纤汤中各大类成分的含量

目的利用紫外分光光度法测定温肺化纤汤中总黄酮、总多酚、总皂苷和总多糖的含量。方法采用硝酸铝显色法测定温肺化纤汤中总黄酮的含量;采用Folin-ciocalteu法测定温肺化纤汤中总多酚的含量;采用香草醛-冰醋酸-高氯酸显色法测定温肺化纤汤中总皂苷的含量;采用硫酸苯酚比色法测定温肺化纤汤中总多糖的含量。结果芦丁在25~200μg·ml-1范围内线性关系良好(r=0.999 0),测得温肺化纤汤中总黄酮含量为(7.37±0.76)mg·ml-1;没食子酸在10~100μg·ml-1范围内线性关系良好(r=0.999 5),测得温肺化纤汤中总多酚含量为(11.83±0.12)mg·ml-1;薯蓣皂苷元在25~200μg·ml-1范围内线性关系良好(r=0.999 7),测得温肺化纤汤中总皂苷含量为(20.75±2.43)mg·ml-1;葡萄糖在10~100μg·ml-1范围内线性关系良好(r=0.999 0),测得温肺化纤汤中总多糖含量为(154.81±3.00)mg·ml-1。结论紫外分光光度法简捷、准确、灵敏度高,可以很好地测定温肺化纤汤中黄酮类、多酚类、皂苷类和多糖类化合物的含量,其结果为温肺化纤汤的新药研发提供了科学依据。 Objective To determine the contents of total flavonoids, total polyphenols, total saponins and total polysaccharides in Wenfei Huaxi Decoction by ultraviolet spectrophotometry. Methods The content of total flavonoids in Wenfei Huaxi Decoction was determined by aluminum nitrate colorimetric method. The content of total polyphenols in Wenfei Huaxi Decoction was determined by Folin-cocalocal method. The content of total polyphenols was determined by vanillin-acetic acid-perchloric acid colorimetry The content of total saponin in the lung fibula soup was determined by phenol-sulfuric acid colorimetry. Results Rutin had a good linearity (r = 0.999 0) in the range of 25 ~ 200μg · ml-1. The content of total flavonoids in Wenfei Huaxian Decoction was (7.37 ± 0.76) mg · ml- The concentration of total polyphenols in Wenfei Huaxian Decoction was (11.83 ± 0.12) mg · ml-1, while that of diosgenin was in the range of 25 ~ 200μg · ml- (R = 0.999 7). The content of total saponins in Wenfei Huaxian Decoction was (20.75 ± 2.43) mg · ml-1. The linearity of glucose in the range of 10 ~ 100μg · ml-1 was good r = 0.999 0). The content of total polysaccharide in Wenfei Huaxian Decoction was (154.81 ± 3.00) mg · ml-1. Conclusion UV spectrophotometry is simple, accurate and sensitive. It can be used to determine the contents of flavonoids, polyphenols, saponins and polysaccharides in Wenfei Huaxian Decoction. The results provide the basis for the development of new drugs for Wenfei Huaxian Decoction The scientific basis.
