目的 :观察重组人白细胞介素 12 (rhIL 12 )对慢性丙型肝炎病毒 (HCV)感染者淋巴细胞增殖的影响。方法 :外周血单个核细胞 (PBMC)分别与植物血凝素 (PHA) ,HCV抗原c2 2、c10 0 3,rhIL 12和抗IL 12抗体孵育 5d后 ,加入胸腺嘧啶核苷(3H TdR) ,然后收集细胞于液闪仪测定每分种脉冲数 (cpm)。结果 :不论是健康对照者 (P <0 .0 2 5 ) ,抑或慢性HCV感染者 (P<0 .0 1) ,rhIL 12在体外可显著增加其淋巴细胞对PHA的增殖反应。同时 ,rhIL 12亦能显著增强HCV感染者对HCV抗原c2 2的增殖反应 (P <0 .0 1)。结论 :rhIL 12可在体外显著增加慢性HCV感染者淋巴细胞的增殖反应 ,该结果为今后使用IL 12治疗这类病人提供了重要的实验根据
Objective: To observe the effect of recombinant human interleukin-12 (IL-12) on lymphocyte proliferation in patients with chronic hepatitis C virus (HCV) infection. Methods: Peripheral blood mononuclear cells (PBMCs) were incubated with phytohemagglutinin (PHA), HCV antigens c2 2, c10 0 3, rhIL 12 and anti-IL 12 antibody for 5 days respectively. Thymidine (3H TdR) The cells are then collected for determination of the number of pulses per minute (cpm) on the flasher. Results: rhIL 12 could significantly increase the lymphocyte proliferation response to PHA in vitro in both healthy controls (P <0.05) and chronic HCV infected patients (P <0.01). At the same time, rhIL 12 also significantly enhanced the HCV infection of c2 2 HCV infection in patients with proliferative response (P <0.01). Conclusion: rhIL 12 can significantly increase the proliferative response of lymphocytes in chronic HCV-infected patients in vitro. This result provides an important experimental basis for the future treatment of patients with IL-12
