目的:探明IL-10和天花粉蛋白的免疫抑制作用,为其临床应用提供理论依据,方法:APC表面分子经荧光染色用FACS分析结果,用~3H-TdR掺入法测定T-细胞增殖,巢式RT-PCR检测B7-1mRNA的表达,结果:IL-10和天花粉蛋白均能显著抑制B7-1分子的表达,两者对ICAM-1的表达未显示抑制作用,IL-10和天花粉蛋白对CD40的表达未见影响,但皆抑制LFA-1的表达,IL-10和天花粉蛋白处理过的APC未检测到B7-1 mRNA的表达,IL-10和天花粉蛋白都能抑制T-细胞的增殖(P<0.01)和IL-2的产生(P<0.01),结论:IL-10和天花粉蛋白抑制APC多种表面分子表达并抑制T-细胞增殖。
Objective: To investigate the immunosuppressive effects of IL-10 and trichosanthin and to provide a theoretical basis for its clinical application. Methods: FACS analysis of APC surface molecules was performed by fluorescent staining. T-cell proliferation was measured by ~ 3H-TdR incorporation, The expression of B7-1 mRNA was detected by nested RT-PCR. Results: IL-10 and trichosanthin significantly inhibited the expression of B7-1, and both of them showed no inhibitory effect on the expression of ICAM-1. IL-10 and trichosanthin But no effect on the expression of CD40, but both inhibited the expression of LFA-1. The expression of B7-1 mRNA was not detected in IL-10 and trichosanthin-treated APCs. Both IL-10 and trichosanthin inhibited T-cell (P <0.01) and IL-2 production (P <0.01). Conclusion: IL-10 and trichosanthin inhibited the expression of various surface molecules of APC and inhibited the proliferation of T-cells.
