目的:评价葡萄膜与结膜色素性肿瘤细胞DNA含量及增殖活性在其良恶性肿瘤鉴别或恶性程度判断上的价值。方法:应用流式细胞术(flowcytometry,FCM)对葡萄膜恶性黑色素瘤(uvealmalignantmelanomas,UMM)、结膜色素痣(conjunctivalnevi,CN)及恶性黑色素瘤(conjunc-tivalmalignantmelanomas,CMM)的DNA含量、增殖指数(proliferativeindex,PI)及S期细胞百分率(s-phasefraction,SPF)进行检测。结果:7例CN均为二倍体,7例CMM及39例UMM的异倍体率分别为57.14%、74.36%。CMM的SPF和PI显著高于CN的SPF(P<0.05)和PI(P<0.01);UMM的DNA指数(DNAindex,DI)、异倍体率与病理类型均表现一定的相关性(P<0.01)。19例UMM的DI与核仁组成区相关嗜银蛋白(argyrophilicnucleolarorganizerregionsassociatedpro-teins,AgNORs)银染面积呈显著正相关(P<0.01)。结论:研究结果提示异倍
Objective: To evaluate the DNA content and proliferative activity of uveal and conjunctival pigmented tumor cells in the differential diagnosis of benign and malignant tumors or the value of malignancy. Methods: The DNA content, proliferative index (VEGF) of uvealmalignant melanomas (UMM), conjunctival nevi (CN) and conjunctival melanoma (CMM) were measured by flow cytometry (FCM) proliferative index (PI) and s-phase fraction (SPF). Results: All 7 CN patients were diploid. The aneuploidy rates of 7 CMM and 39 UMM were 57.14% and 74.36% respectively. The SPF and PI of CMM were significantly higher than that of CN (P <0.05) and PI (P <0.01). The DNA index (DI) and aneuploidy rates of UMM were correlated with pathological types (P <0.01). There was a significant positive correlation between DI of 19 cases of UMM and the area of silver staining of argyrophilic nucleolar organizers associated with proteins (AgNORs) (P <0.01).
