静脉注射酵母多糖激活的血清(ZAS)或甲酰甲硫氨酰白氨酰苯丙氨酸(FMLP一种合成趋化性肽)可在大白鼠产生一短暂但严重的嗜中性白细胞减少。来自补体的肽和FMLP均可引起嗜中性白细胞膜的变化从而导致白细胞附着于血管壁和凝聚,凝聚的嗜中性白细胞被羁留在肺毛细血管床。因此,嗜中性白细胞减少是附着和凝聚的联合所引起。本文研究FMLP和ZAS在体外对多形核白细胞的凝聚作用以及一种体外凝聚模型能为研究作用于白细胞功能的抗炎药物的构-效关系,提供一有用的方法。
Intravenous zymosan activated serum (ZAS) or formylthionyl leucylphenylalanine (FMLP, a synthetic chemotactic peptide) produced a transient but severe neutropenia in rats. Peptides from both complement and FMLP can cause neutrophil membrane changes leading to attachment of white blood cells to the vessel wall and coagulation, and coagulated neutrophils are trapped in the pulmonary capillary bed. Therefore, neutropenia is caused by a combination of attachment and aggregation. This study of FMLP and ZAS agglutination of polymorphonuclear leukocytes in vitro and an in vitro cohesion model provide a useful method for studying the structure-activity relationship of anti-inflammatory drugs acting on leukocytes.
