目的研究卡介菌多糖核酸(BCG-PSN)对PMA刺激的U937细胞分泌炎性因子TNF-αI、L-1β的影响。方法人髓系白血病细胞U937用PMA诱导成熟后分5组:正常对照组、LPS组、高剂量、中剂量、低剂量BCG-PSN组,培养24 h后收集上清,ELISA测定细胞培养上清中TNF-α、IL-1β的浓度。结果 BCG-PSN刺激PMA诱导的U937产生TNF-α明显增多,与正常对照组相比,差异有统计学意义(P<0.05)。BCG-PSN刺激PMA诱导的U937产生的IL-1β与正常对照组相比,差异无统计学意义。结论 BCG-PSN可以刺激PMA诱导的U937细胞产生TNF-α,对U937产生的IL-1无明显的影响。
Objective To investigate the effects of BCG-PSN on inflammatory cytokines TNF-αI and L-1β secreted by PMA-stimulated U937 cells. Methods Human myeloid leukemia cell line U937 was induced to mature by PMA and divided into 5 groups: normal control group, LPS group, high dose, middle dose and low dose BCG-PSN group. After cultured for 24 h, the supernatant was harvested and the cell culture supernatants In the concentration of TNF-α, IL-1β. Results BCG-PSN stimulated PMA-induced U937 TNF-α production increased significantly, compared with the normal control group, the difference was statistically significant (P <0.05). BCG-PSN stimulated PMA-induced U937 produced IL-1β compared with the normal control group, the difference was not statistically significant. Conclusion BCG-PSN can stimulate PMA-induced U937 cells produce TNF-α, U937 produced IL-1 no significant effect.
