根据药理学特性α_1肾上腺素受体(α_1-AR)曾被分成α_(1A)-和α_(1B)-两种亚型;据此我们曾系统观察这两种亚型在大鼠血管中的分布、调节及信号转导等方面的差异,提出在大鼠主动脉和肾动脉中的功能性α_1-AR分别属于α_(1B)-和α_(1A)-AR亚型。随着研究的不断深入特别是分子生物学技术的应用,发现α_1-AR实际上还包含第三种亚型,即α_(1D)-AR亚型。因此有必要对我们曾报道的根据两种分型所观察到的大鼠血管的α_1-AR分型重新进行研究。本文采用离体血管收缩功能实验和逆转录-多聚酶链反应(RT-PCR)方法,研究3种α_1-AR亚型在大鼠主动脉和肾动脉的分布情况及其功能意义。
According to the pharmacological properties of α_1 adrenergic receptor (α_1-AR) has been divided into α 1A (1A) and α 1B (1B) - two subtypes; accordingly, we have systematically observed these two subtypes in rat blood vessels Distribution, regulation and signal transduction, we propose that functional alpha 1-AR in rat aorta and renal arteries belong to α 1B and α 1A subtypes, respectively. With the deepening of research, especially the application of molecular biology techniques, it was found that α_1-AR actually contains the third subtype, that is, the α_ (1D) -AR subtype. Therefore, it is necessary to re-study the α_1-AR type of rat blood vessels we have reported based on the two subtypes. In this study, we used ex vivo vasoconstriction test and reverse transcriptase-polymerase chain reaction (RT-PCR) to study the distribution of three α1-AR subtypes in the aorta and renal arteries of rats and their functional significance.
