以西洋参种胚诱导愈伤组织上的体细胞胚(胚状体)为试材,采用离体培养方法,研究胚状体自身发育阶段及用于成熟、萌发生长的培养基的组成因子对其成熟萌发的影响,以期确定最佳的胚状体成熟培养基和萌发培养基。结果表明:发育阶段为鱼雷形的胚状体,是进行成熟培养的最佳材料;胚状体成熟的最佳培养基是0.5M+0.4mg/L 6-BA+2mg/L NAA+5mg/L GA3+1%AC;在此培养基上胚状体生长变大,子叶生长展开,胚芽、胚根肉眼可见;成熟培养基中植物生长调节物质在活性炭作用下缓慢释放并作用于胚状体,达到形成大量正常、健壮、同步胚状体的目的;胚状体萌发的最佳培养基是0.5MA+0.4mg/L 6-BA+2mg/L NAA+5mg/L GA3;在此培养基上胚状体萌发生长,胚芽伸长真叶展开。
