目的:比较胰岛素样生长因子1(insulinlikegrowthfactor1,IGF1)和表皮生长因子(epidermalgrowthfactor,EGF)对高浓度葡萄糖的腹膜间皮细胞毒性作用的影响。方法:体外培养人腹膜间皮细胞,在上清中加入83.3mmol·L-1葡萄糖和不同浓度IGF1或EGF,培养24、48和72h后,测定3H胸腺嘧啶核苷(3HTdR)掺入率和上清中乳酸脱氢酶(LDH)水平。结果:50μg·L-1的IGF1作用72h后,83.3mmol·L-1葡萄糖溶液中培养间皮细胞的3HTdR掺入率较高糖抑制组增加(75±15)%(P<0.01),作用24h,上清中LDH水平较高糖抑制组减少(34±8)%(P<0.01);EGF对高糖细胞毒性无明显影响(P>0.05)。结论:IGF1能拮抗高浓度葡萄糖对人腹膜间皮细胞的毒性作用。
Objective: To compare the effects of insulinlike growth factor1 (IGF1) and epidermal growth factor (EGF) on the peritoneal mesothelial cell toxicity of high glucose. Methods: Human peritoneal mesothelial cells were cultured in vitro, and 83.3mmol·L-1 glucose and different concentrations of IGF1 or EGF were added into the supernatant. After 24, 48 and 72h incubation, 3HThymidine (3H TdR incorporation rate and lactate dehydrogenase (LDH) level in the supernatant. Results: After treated with 50μg · L-1 IGF1 for 72h, the 3HTdR incorporation rate of mesangial cells in 83.3mmol·L-1 glucose solution increased (75 ± 15)% (P (P <0.01). The level of LDH in the supernatant was decreased by 34 ± 8% (P <0.01), while the level of LDH in the supernatant was no significant difference (P> 0.05) . Conclusion: IGF 1 can antagonize the high concentration of glucose on the human peritoneal mesothelial cell toxicity.
