目的:探讨佛手组织培养中褐变的控制方法。方法:以佛手顶芽及带节茎为材料,开展了不同基本培养基、取材时间、激素种类和褐变抑制剂对佛手初代培养中外植体褐变的影响研究。结果:佛手初代培养选用1/2MS为基本培养基并附加1.0 mg/L 6-BA、0.2 mg/L NAA、1.0 mg/L ZT和3 g/L PVP,1~3月份取材,是控制褐变的有效措施。结论:适宜的培养基组分和抗褐变剂能明显减轻佛手组织培养中的褐变,提高组培成功率,初步建立佛手组织培养体系。
Objective: To explore the method of controlling browning in tissue culture of bergamot. Methods: The effects of different basal mediums, time, hormones and inhibitors of browning on the browning of explants during the initial culture of bergamot were studied using the top buds and stems. Results: The initial culture of bergamot picked 1/2 MS as basic medium and supplemented with 1.0 mg / L 6-BA, 0.2 mg / L NAA, 1.0 mg / L ZT and 3 g / Change effective measures. Conclusion: The appropriate medium components and anti-browning agents can significantly reduce the browning of bergamot tissue culture, improve the success rate of tissue culture, and establish the tissue culture system of bergamot.
