核苷酸切除修复系统在维持基因组稳定、修复致癌因素所致的损伤及抗癌过程中发挥着重要作用。研究证实基因多态性可能引起氨基酸改变,然后导致表达的蛋白质异常,进而影响DNA修复能力。因此,基因多态性与肿瘤的发病风险和遗传易患性密切相关。同时在化疗过程中,其与肿瘤敏感性、中位生存时间等显著相关。切除修复交叉互补基因2(ERCC2)在核苷酸切除修复系统中发挥着重要作用,现就ERCC2基因多态性与胃癌的易患性以及化疗药物之间的关系予以综述。
Nucleotide excision repair system plays an important role in maintaining genomic stability, repairing damage caused by carcinogenic factors and anti-cancer process. Studies have confirmed that genetic polymorphisms may cause amino acid changes, and then lead to abnormal expression of the protein, thereby affecting DNA repair capacity. Therefore, genetic polymorphisms are closely related to the risk of developing tumors and genetic predisposition. At the same time in the course of chemotherapy, it has significant correlation with tumor sensitivity, median survival time and so on. Resection and repair of cross-complementary gene 2 (ERCC2) play an important role in the nucleotide excision repair system. The relationship between ERCC2 gene polymorphism and susceptibility to gastric cancer and chemotherapeutic drugs are reviewed.
