目的:探讨戊己丸不同配伍时,代表成分胆汁排泄的变化。从胆汁排泄的角度解释戊己丸的配伍机制。方法:采用正交设计得到9个戊己丸复方,加上9个单味药,共18个复方。于十二指肠给药后不同时间点取胆汁,用LC-MS测定代表性成分的胆汁浓度,以此计算胆汁排泄率,药-时曲线下面积,并对结果进行方差和极差分析。结果:吴茱萸与小檗碱、巴马汀胆汁排泄呈负相关;白芍与小檗碱、巴马汀胆汁排泄呈正相关;黄连、白芍与吴茱萸碱、吴茱萸次碱胆汁排泄呈负相关;并且得到了针对各代表成分胆汁排泄的最优配伍比例。结论:吴茱萸抑制黄连代表性成分胆汁排泄;白芍促进黄连代表性成分胆汁排泄;黄连、白芍抑制吴茱萸代表性成分胆汁排泄;黄连、吴茱萸抑制白芍代表性成分的胆汁排泄。
OBJECTIVE: To investigate the changes of bile excretion of different compositions of Wuji Wan. Explain the Compatibility Mechanism of Wuji Pills from the Angle of Bile Excretion. Methods: Using orthogonal design, we obtained 9 compounds of Wuji Pills, plus 9 single herbs, a total of 18 compounds. Bile was taken at different time points after duodenum administration, and the bile concentration of representative components was determined by LC-MS. The bile excretion rate and the area under the drug-time curve were calculated, and the results were analyzed for variance and range. Results: There was a negative correlation between Evodia rutaecarpa and berberine and palmatine bile excretion. There was a positive correlation between pearsis and berberine and palmatine bile excretion. Coptis chinensis and Radix Paeoniae Alba were negatively correlated with evodiamine and rutaecarpine bile excretion. The optimum proportion of bile excretion for all representative components was obtained. Conclusion: Fructus Evodiae can inhibit the bile excretion of the representative constituents of Coptis chinensis; white peony root promotes the bile excretion of the representative constituents of Coptis chinensis; Coptis chinensis and Radix Paeoniae Alba can inhibit the excretion of biliary excretion of the components of Fructus Evodiae; Coptis chinensis and Fructus Evodiae inhibit the biliary excretion of the representative components of Radix Paeoniae rubra.
