目的:探讨人类疱疹病毒6型(HHV6)与血液病患者巨噬细胞集落刺激因子(MCSF)异常表达的关系。方法:以卵白素生物素复合物(ABC)免疫酶标和逆转录聚合酶链反应(RTPCR)方法检测30例血液病患者骨髓单个核细胞体外感染HHV6后MCSF和MCSFmRNA的表达。结果:HHV6感染组MCSF阳性细胞百分率、积分值和MCSFmRNA检出率分别为36.9%±24.9%,45.9±31.4和833%,显著高于对照组(20.3%±19.4%,25.3±24.2,50.0%)(P<0.001);其中急性白血病组和慢性粒细胞白血病组MCSF表达升高幅度较大。MCSF主要分布于粒细胞和单核细胞的胞膜和胞浆。结论:体外感染HHV6可以使血液病患者骨髓单个核细胞MCSF表达升高
Objective: To investigate the relationship between human herpes virus type 6 (HHV6) and abnormal expression of macrophage colony-stimulating factor (MCSF) in patients with hematological diseases. Methods: Bone marrow mononuclear cells from 30 patients with hemopathy were tested for in vitro infection of H-CSF with HvV-6 by immunoblot and reverse transcription polymerase chain reaction (RT-PCR) of avidin-biotin complex (ABC) M CSF mRNA expression. Results: The positive rates of M-CSF positive cells in HHV6 infection group were 36.9% ± 24.9%, 45.9 ± 31.4 and 833% respectively, which were significantly higher In the control group (20.3% ± 19.4%, 25.3 ± 24.2, 50.0%) (P <0.001). The expression of M-CSF in acute leukemia group and chronic myeloid leukemia group High amplitude. M-CSF is mainly distributed in the cell membrane and cytoplasm of granulocytes and monocytes.
