研究内皮素-1(ET-1)在猫延髓头侧腹外侧区(rVLM)对心血管活动的影响.方法:采用脑立体定向和脑内微量注射技术.结果:rVLM内微量注射 ET-1(4μmol·L-1, 0.5μL)后,平均动脉压(MAP)升高(3.7±1.3)kPa,心率增加(29±7)beats·min-1,肾神经活动增加 45%±10%,量-效呈正相关.双颈部迷走神经切断后不影响上述结果.静脉预先给予酚妥拉明 5 mg·kg-1能显著抑制ET-1的升压作用.血浆精氨酸升压素(Arg)含量由(12.4± 6. 5) ng· L-1增加到(70.3± 24. 2)ng·L-1,并与MAP变化显著相关,使缺血心脏心律紊乱(HRhD),HRhD出现在结扎后(4.8±2.9)min,评分为 4.4±1.6,与对照组差异显著.结论:rVLM内注射ET-1影响了心血管和交感神经活动的中枢调节。
To investigate the effect of endothelin-1 (ET-1) on cardiovascular activity in the rostral ventrolateral medulla (rVLM). Methods: Brain stereotactic and intracerebral microinjection techniques were used. RESULTS: Mean arterial pressure (MAP) increased (3.7 ± 1.3) kPa and heart rate increased (29 ± 7)% after microinjection of ET-1 (4μmol·L-1, 0.5μL) into rVLM min-1, renal nerve activity increased by 45% ± 10%, the amount of - effect was positively correlated. Two-neck vagus nerve cut off does not affect the above results. Pretreatment of intravenous phentolamine 5 mg · kg-1 significantly inhibited the ET-1 vasopressor effect. The content of Arginine in plasma increased from (12.4 ± 6.5) ng · L-1 to (70.3 ± 2. 2) ng · L-1, and was significantly correlated with the change of MAP. The HRhD and HRhD were significantly higher than those of the control group (4.8 ± 2.9) min after the ligation, with a score of 4.4 ± 1.6. Conclusions: Intravenous injection of ET-1 into rVLM affects central regulation of cardiovascular and sympathetic activity.
