目的 :研究脑络欣通对脑缺血再灌注模型鼠神经细胞凋亡及P5 3蛋白表达的影响。方法 :采用饥饿、劳累、高脂饮食等多因素复制大鼠气虚血瘀证模型 ,然后用线栓法阻断大鼠左侧大脑中动脉 2h ,复制局灶性脑缺血再灌注模型 ,于再灌注 1,3d ,用TUNEL法和免疫组化法分别检测脑缺血半暗带凋亡细胞和P5 3蛋白表达。结果 :与模型组比较 ,脑络欣通组脑缺血半暗带的凋亡细胞和P5 3蛋白表达显著减少 (P <0 .0 5 )。结论 :抑制P5 3蛋白的表达是脑络欣通抗脑缺血再灌注后神经细胞凋亡的机制之一。
Objective : To study the effect of Naoluoxintong on apoptosis of neurons and expression of P53 protein in rat model of cerebral ischemia-reperfusion. METHODS: Rat models of qi deficiency and blood stasis syndrome were duplicated by various factors including starvation, fatigue, and high-fat diet. The left middle cerebral artery was then blocked for 2 h by a wire embolus method and the model of focal cerebral ischemia/reperfusion was replicated. After reperfusion for 1 and 3 days, the apoptotic cells and P53 protein expression in the penumbra of ischemic penumbra were detected by TUNEL and immunohistochemistry, respectively. RESULTS: Compared with the model group, the expression of apoptotic cells and P53 protein in the cerebral ischemic penumbra of the Naoluoxintong group was significantly reduced (P < 0.05). Conclusion : Inhibition of P53 protein expression is one of the mechanisms of Naoluoxintong against neuronal apoptosis after cerebral ischemia-reperfusion.
