目的:黄芪注射液与TAM合用对雌激素受体阳性人乳腺癌MCF-7细胞增殖凋亡的影响。方法:实验分为空白对照组,TAM对照组和与TAM合用5个不同剂量的黄芪注射液组。MTT法检测增殖,流式细胞仪分析细胞周期,PI染色检测凋亡率,DAN ladder观察凋亡。结果:与TAM合用,各剂量黄芪注射液均呈现抑制MCF-7细胞增殖的效应(P<0.05),随着作用时间延长,72 h其效应呈剂量依赖性。高剂量黄芪注射液作用72 h诱导MCF-7细胞凋亡。与TAM合用24 h,2×10-1g/mL剂量组阻滞细胞周期于S期(P<0.05),2×10-2g/mL剂量组阻滞细胞周期于G0/G1期(P<0.05)。结论:与TAM合用,黄芪可抑制雌激素受体阳性的乳腺癌细胞增殖,一定剂量范围内可诱导其凋亡,阻滞细胞的有丝分裂于S或G2/M期。
Objective: To investigate the effects of Huangqi injection combined with TAM on proliferation and apoptosis of estrogen receptor positive human breast cancer MCF-7 cells. Methods: The experiment was divided into blank control group, TAM control group and 5 different doses of astragalus injection group combined with TAM. Cell proliferation was detected by MTT assay, cell cycle was analyzed by flow cytometry, apoptosis rate was detected by PI staining, apoptosis was observed by DAN ladder. Results: With the combination of TAM and Astragalus mongholicus, the effects of astragalus mongholicus injection on proliferation of MCF-7 cells were inhibited (P <0.05). With the prolongation of action time, the effect was dose-dependent at 72 h. High-dose Astragalus injection induced 72-h apoptosis in MCF-7 cells. The cells in 2 × 10-1g / mL dose group blocked the cell cycle in S phase (P <0.05), and the cell cycle in 2 × 10-2g / mL dose blocked the G0 / G1 phase (P <0.05) ). Conclusion: Combined with TAM, Astragalus can inhibit estrogen receptor-positive breast cancer cell proliferation, a certain dose range can induce apoptosis, arrest cell mitosis in the S or G2 / M phase.
