目的探讨孕期叶酸干预对幼年仔鼠神经反射行为的影响。方法选用SPF级成年雌性SD大鼠48只,采用孕期饲料干预的方法,随机将雌鼠分为4组:叶酸缺乏组(D组,0.1 mg/kg)、叶酸正常组(N组,2.1 mg/kg)、孕前孕后叶酸短期补充组(S组孕前3.5 mg/kg,孕后2.1 mg/kg)及孕前孕后叶酸全程补充组(L组,3.5 mg/kg),每组12只。各组均从不同窝中随机选取30只子代鼠,于子代出生后第7~14天进行触须定位反射、伸肌交叉反射及感官反射检测,比较各组仔鼠出现神经反射行为的阳性率。结果各组仔鼠神经反射行为的阳性率均随着日龄的增加而逐渐增加,触须定位、伸肌交叉及耳肌反射在实验后期所有子鼠均表现为阳性。L组和S组神经反射出现较早,D组出现最晚。组间比较各神经反射均表现为L组和S组阳性率较高,其次为N组,D组最低。结论孕期叶酸缺乏会延迟仔鼠神经反射出现的时间,叶酸补充可改善由叶酸缺乏引起的仔鼠神经反射延迟,提示孕期叶酸干预对仔鼠神经功能的成熟程度有一定的影响。
Objective To investigate the effect of folic acid intervention during pregnancy on the neurological reflexes of young offspring. Methods Forty-eight adult Sprague-Dawley (SD) female Sprague-Dawley rats were randomly divided into 4 groups: folic acid deficiency group (D group, 0.1 mg / kg), folic acid normal group / kg), folic acid supplementation group (3.5 mg / kg before pregnancy, 2.1 mg / kg after pregnancy) and folic acid supplementation group (L group, 3.5 mg / kg) Thirty male mice were randomly selected from different litters in each group. Targeted locomotion reflex, extensor muscle cross-reflex and sensory reflex detection were performed on the 7th to 14th day after birth. The positive reflex behavior of the pups was compared rate. Results The positive rate of neural reflexes in each group increased gradually with the increase of age. The location of tentacles, extensor cross and ear muscle reflex were all positive in the later stage of experiment. Neurological reflexes appeared earlier in group L and group S, and appeared late in group D. Comparisons among groups showed that all neurological reflexes had the highest positive rate in L group and S group, followed by N group and the lowest in D group. Conclusions Folic acid deficiency during pregnancy can delay the time of neurological reflex in pups. Folic acid supplementation can improve the neurological reflex delay of pups induced by folic acid deficiency, suggesting that folic acid intervention during pregnancy may have some influence on the neural function maturation of pups.
