目的 :研究eIF_5A的亚细胞定位 ,为了解其生物学功能提供线索。方法 :借助免疫荧光技术和GFP标签技术 ,用激光共聚焦显微镜检测eIF_5A的亚细胞定位。结果 :免疫荧光检测结果表明 ,eIF_5A定位于细胞质中 ;GFP_eIF_5A瞬时转染后 ,起初eIF_5A为全细胞分布 ,以后逐渐转移到细胞质中。第 5 0位赖氨酸突变为丙氨酸后的eIF_5A则分布于整个细胞。结论 :eIF_5A具有核质穿梭功能 ,它的亚细胞定位是个动态变化过程 ,hypusine可影响其亚细胞定位 ,进而可能影响其功能的发挥。
Objective: To study the subcellular localization of eIF_5A and provide clues for understanding its biological function. METHODS: Subcellular localization of eIF_5A was detected by laser confocal microscopy with immunofluorescence and GFP labeling. Results: The result of immunofluorescence showed that eIF_5A localized in the cytoplasm. After transient transfection of GFP_eIF_5A, eIF_5A initially distributed in the whole cell and then gradually transferred to the cytoplasm. The eIF_5A after mutation of lysine at position 50 to alanine is distributed throughout the cell. Conclusion: eIF_5A has the function of nucleoplasm shuttle, its subcellular localization is a dynamic process, hypusine can affect its subcellular localization, which may affect its function.
