目的:探讨白藜芦醇(resveratrol,Res)对过表达EGFR的Ⅲ型突变体(EGFRvⅢ)的U87人脑胶质瘤细胞(U87-EGFRvⅢ)的生长抑制作用。方法:将不同浓度的Res作用于U87-EGFRvⅢ或U87细胞,用MTT法检测Res对细胞的生长抑制作用,流式细胞仪检测细胞凋亡作用,免疫印迹法检测Res作用后细胞内EGFRvⅢ的表达情况。结果:经不同浓度Res处理后的U87-EGFRvⅢ细胞和U87细胞均显示抑制细胞生长的作用(P<0.05);流式细胞仪显示不同浓度Res对U87-EGFRvⅢ有促凋亡作用,显著高于对照组(P<0.05),100μmol/L和200μmol/L Res的48 h的细胞凋亡率分别为20.1%±0.8%、46.9%±1.2%;免疫印迹法显示Res作用后U87-EGFRvⅢ细胞内的EGFRvⅢ的表达量显著下降。结论:Res可诱导U87-EGFRvⅢ细胞凋亡、抑制其细胞增殖,并可使细胞内的EGFRvⅢ表达下降。
AIM: To investigate the inhibitory effect of resveratrol on the growth of U87 human glioma cells (U87-EGFRvⅢ) over-expressing EGFRv Ⅲ. Methods: Different concentrations of Res were administered to U87-EGFRvⅢ or U87 cells. The growth inhibition of Res was detected by MTT assay. The apoptosis of cells was detected by flow cytometry. The expression of EGFRvⅢ was detected by Western blotting Happening. Results: U87-EGFRvIII cells and U87 cells treated with different concentrations of Res showed inhibitory effects on cell growth (P <0.05). Flow cytometry showed that different concentrations of Res had pro-apoptotic effects on U87-EGFRvIII, which were significantly higher than that of U87- The apoptosis rate of U87-EGFRvⅢ cells treated with Res for 48 hours after resveratrol treatment was 20.1% ± 0.8% and 46.9% ± 1.2% respectively in the control group (P <0.05) The expression of EGFRvIII decreased significantly. Conclusion: Res can induce apoptosis of U87-EGFRvIII cells, inhibit its proliferation and decrease the expression of EGFRvIII in cells.
