目的:探讨血清贮存对检测血清T-PSA、F-PSA及F-PSA%(F-PSA/T-PSA)结果的影响。方法:应用全自动化学发光分析仪检测不同温度下贮存的血清F-PSA、T-PSA及F-PSA%值,并对各组数值结果进行比较分析。结果:①血清贮存在4℃和室温(22~26℃)时,F-PSA较T-PSA不稳定;室温下4 h后F-PSA值下降为(0.392±0.246)μg/L,较初始值比较有显著性变化(P<0.01),而T-PSA、F-PSA%分别下降为(1.522±1.085)μg/L和(25.03±5.94)%,无显著性变化(P>0.05)。室温下8 h,T-PSA、F-PSA分别降为(1.513±1.083)、(0.389±0.247)μg/L,与初始值比较均有显著性差异(P<0.05,P<0.01)。血清贮存在4℃,2 d后T-PSA、F-PSA及F-PSA%值分别降为(9.418±7.965)、(2.168±1.558)μg/L和(26.6±6.63)%,与初始值比较,均发生显著性变化(P均<0.05);贮存1周后三者结果分别为(9.203±7.736)μg/L、(2.047±1.478)μg/L和(25.64±6.56)%(P均<0.01)。血清贮存在-40℃时,4周后T-PSA、F-PSA及F-PSA%值分别为(4.532±4.393)、(1.178±1.034)μg/L和(24.45±8.81)%。②血清在-40℃反复冻融3次后检测T-PSA和F-PSA值分别为(5.982±5.314)μg/L和(1.341±1.029)μg/L,与初始值比较,无明显变化(P>0.05)。结论:血清贮存温度不同对血清T-PSA、F-PSA及F-PSA%的检测有不同程度的影响,且F-PSA较T-PSA变化大,F-PSA%相对稳定;血清贮存在-40℃,T-PSA和F-PSA至少可稳定1个月;血清反复冻融对PSA检测无影响。
Objective: To investigate the effect of serum storage on the detection of serum T-PSA, F-PSA and F-PSA / T-PSA. Methods: The F-PSA, T-PSA and F-PSA% of serum stored at different temperatures were detected by automatic chemiluminescence analyzer. The numerical results of each group were compared and analyzed. Results: (1) The F-PSA was not stable when stored at 4 ℃ and room temperature (22-26 ℃). The F-PSA decreased to (0.392 ± 0.246) μg / L after 4 h at room temperature, (P <0.01). However, the levels of T-PSA and F-PSA decreased to (1.522 ± 1.085) μg / L and (25.03 ± 5.94)% respectively, with no significant change (P> 0.05). The levels of T-PSA and F-PSA decreased to (1.513 ± 1.083) and (0.389 ± 0.247) μg / L at 8 h at room temperature, respectively, which were significantly different from the initial values (P <0.05, P <0.01). The serum T-PSA, F-PSA and F-PSA% values were reduced to (9.418 ± 7.965), (2.168 ± 1.558) μg / L and (26.6 ± 6.63)% after 2 days of storage at 4 ℃, (P <0.05). After one week of storage, the three results were (9.203 ± 7.736) μg / L, (2.047 ± 1.478) μg / L and (25.64 ± 6.56)% <0.01). The serum T-PSA, F-PSA and F-PSA% values were (4.532 ± 4.393), (1.178 ± 1.034) μg / L and (24.45 ± 8.81)% after 4 weeks of storage at -40 ℃. ② The serum T-PSA and F-PSA values were (5.982 ± 5.314) μg / L and (1.341 ± 1.029) μg / L, respectively, after repeated freeze-thaw cycles at -40 ℃ P> 0.05). CONCLUSIONS: The different serum storage temperatures have different effects on the detection of serum T-PSA, F-PSA and F-PSA, and the changes of F-PSA are bigger than T-PSA and the F- At 40 ℃, T-PSA and F-PSA can be stable for at least 1 month; serum freeze-thaw repeatedly has no effect on PSA test.
