目的 :探讨全反式维甲酸 (all transretinoicacid ,ATRA)诱导肿瘤细胞分化的分子机制。方法 :采用Southern和Northern杂交方法对本室用ATRA诱导人肺腺癌细胞GLC 82 ,经消减杂交所构建的cDNA消减文库进一步鉴定 ;序列分析后用RT PCR方法研究胎儿组织中的表达情况。结果 :分离到一为ATRA激活表达的基因—PIGB1,该基因与人PIG B (phosphatidylinositolglycanofcomplementationclassB)基因高度同源 ,在多种胎儿组织中均有表达。结论 :PIGB1基因可能参与ATRA诱导肿瘤细胞分化过程
Objective: To explore the molecular mechanism of all-trans retinoic acid (ATRA)-induced tumor cell differentiation. METHODS: Southern and Northern blotting methods were used to identify human lung adenocarcinoma cell line GLC82 by ATRA in the laboratory and further identified by cDNA subtracted library constructed by subtractive hybridization. RTPCR method was used to study the expression of the human fetal tissue after sequence analysis. RESULTS: A gene that was activated and expressed by ATRA, PIGB1, was isolated. This gene is highly homologous to human PIG B (phosphatidylinositolglycanofcomplementationclassB) gene and expressed in various fetal tissues. Conclusion : PIGB1 gene may be involved in ATRA-induced differentiation of tumor cells
