目的探讨白介素8(IL-8)基因表达在原发性乳腺癌组织中的表达及其与预后的关系。方法收集55例原发性乳腺癌患者的临床病理及随访资料,中位随访时间90个月。采用实时定量PCR方法检测5 5例乳腺癌IL-8的mRNA相对表达值,免疫组化EnVision法检测乳腺癌组织中雌激素受体(ER),孕激素受体(PR),C-erbB-2的表达情况;分析患者年龄,淋巴结转移,ER,PR,C-erbB-2和IL-8的相对表达及治疗方式等与预后的关系。运用受试者操作特性(ROC)曲线评价IL-8预测疾病复发的能力。结果 ROC发现IL-8相对表达值取5时,能获得最佳的预后能力。单因素生存分析表明:淋巴结有转移,未进行内分泌治疗,ER阴性,IL-8相对表达高(比值≥5)时乳腺癌患者的无疾病生存期(RFS)较短(P=0.0029,0.035,0.026及0.0027)。多因素生存分析表明:淋巴结和IL-8相对表达倾向为预测乳腺癌患者RFS的独立因素(P=0.017及P=0.094)。结论乳腺肿瘤内IL-8相对表达量可能是预测原发性乳腺癌患者术后复发转移的参考因素。
Objective To investigate the expression of interleukin-8 (IL-8) gene in primary breast cancer and its relationship with prognosis. Methods The clinical data of 55 patients with primary breast cancer were collected and followed up for a median of 90 months. The mRNA expression of IL-8 in breast cancer was detected by real-time PCR. The expression of ER, PR, 2, and analyze the relationship between prognosis and age, lymph node metastasis, relative expression of ER, PR, C-erbB-2 and IL-8 and treatment methods. The ability of IL-8 to predict disease recurrence was evaluated using a receiver operating characteristic (ROC) curve. Results ROC found that when the relative expression value of IL-8 was taken as 5, the best prognosis ability was obtained. Univariate survival analysis showed that patients with breast cancer had shorter disease-free survival (RFS) when lymph node metastasis, endocrine therapy, ER negative, and IL-8 relative expression were higher (P = 0.0029,0.035, 0.026 and 0.0027). Multivariate survival analysis showed that the relative expression of lymph nodes and IL-8 was an independent predictor of RFS in breast cancer patients (P = 0.017 and P = 0.094). Conclusion The relative expression of IL-8 in breast tumor may be the reference factor to predict the recurrence and metastasis of primary breast cancer patients.
