目的研究神经肽Y(NPY)和神经元特异性烯醇酶(NSE)在戊四氮(PTZ)致癫痫(EP)大鼠不同脑区中的变化,探讨NPY与EP的关系及其对大脑神经元的损坏。方法以SD大鼠50只进行试验。对照组(A组)注射生理盐水。实验组腹腔注射PTZ 50 mg/kg。据EP发作分级,≤1级视为B组;2级以上于发作后0、6、24 h(C、D和E组)断头取脑,分别取中脑、海马、纹状体和额叶等部位,用RIA法测定NPY,EIA法测定NSE;以免疫组化染色法观察海马的NPY和NSE表达。结果 EP发作组各部位的NPY含量均高于A组;EP后各部位及血清的NSE均明显高于A组,表明有脑组织损害。结论 NPY与EP发作有密切的关系。EP发作后NSE明显升高,尤其是海马区,表明EP发作会造成大脑组织损坏。
Objective To investigate the changes of neuropeptide Y (NPY) and neuron specific enolase (NSE) in different brain regions of PTZ-induced epileptic rats (EPZ), and to explore the relationship between NPY and EP and its effect on brain Neuronal damage. Methods Fifty SD rats were tested. The control group (group A) received normal saline. Experimental group intraperitoneal injection of PTZ 50 mg / kg. After the onset of EP NSE significantly increased, especially in the hippocampus, indicating that the onset of EP will cause damage to brain tissue.
