目的观察扶正消癌方对Lewis肺癌模型小鼠肿瘤生长的抑制作用及其机制。方法选择60只C57BL/6J小鼠右前肢腋下接种Lewis肺癌细胞建立Lewis肺癌小鼠模型,随机分为荷瘤模型组、环磷酰胺(CTX)组、中药预防组、综合组,每组15只。CTX组接种72h后一次性腹腔注射CTX溶液0.2ml(4mg/ml),中药预防组接种前5天开始灌胃扶正消癌方0.4ml(2g/ml),综合组在CTX组基础上灌胃扶正消癌方药0.4ml(2g/ml),荷瘤模型组给予0.4ml生理盐水灌胃;每天灌胃1次,至造模后12天。观察各组小鼠生存状态及移植瘤生长情况,称量瘤重、胸腺重,计算抑瘤率、胸腺指数。免疫组化法检测瘤组织细胞黏附因子CD44的表达。结果与荷瘤模型组比较,各组瘤重均显著降低(P<0.01),综合组瘤重显著低于CTX组(P<0.01),CTX组、中药预防组、综合组抑瘤率分别为83.97%、55.71%、87.23%。与荷瘤模型组比较,CTX组和综合组胸腺重均显著降低(P<0.05或P<0.01),CTX组胸腺指数显著降低(P<0.01);与CTX组比较,中药预防组胸腺重量显著增加(P<0.01),综合组胸腺指数显著增加(P<0.05)。综合组CD44表达程度最低,与CTX组比较差异有统计学意义(P<0.05)。结论扶正消癌方可以协同化疗药物,提高肿瘤细胞对免疫系统及抗癌药物的敏感性,下调或逆转Lewis肺癌模型小鼠黏附因子CD44表达,发挥抑瘤和抗转移作用。
Objective To observe the inhibitory effect of Fuzheng XiaoJian Fang on tumor growth in Lewis lung carcinoma model mice and its mechanism. Methods Sixty Lewis lung cancer cells were inoculated into the right forelimb of Lewis lung carcinoma in 60 C57BL / 6J mice. Lewis lung cancer mice were randomly divided into three groups: tumor-bearing model group, cyclophosphamide (CTX) group, prophylaxis group, and combination group only. The CTX group received intraperitoneal injection of 0.2 ml (4 mg / ml) of CTX solution 72 h after inoculation, and 0.4 ml (2 g / ml) Fuzheng Xiao Fang Fang 0.4ml (2g / ml), tumor-bearing model group given 0.4ml saline gavage; gavage once daily until 12 days after modeling. The survival status of the mice in each group and the growth of the transplanted tumor were observed. The tumor weight and the weight of the thymus were weighed, and the tumor inhibition rate and the thymus index were calculated. Immunohistochemistry was used to detect the expression of CD44 on tumor cells. Results Compared with the tumor-bearing model group, the tumor weight in each group was significantly decreased (P <0.01), the tumor weight in the combined group was significantly lower than that in the CTX group (P <0.01), and the tumor inhibition rates in the CTX group and the traditional Chinese medicine prophylaxis group were 83.97%, 55.71%, 87.23%. Compared with the CTX group, the thymus weight in the CTX group and the combined group were significantly decreased (P <0.05 or P <0.01), and the thymus index in the CTX group was significantly decreased (P <0.01). Compared with the CTX group, the thymus weight in the TCM prevention group was significantly (P <0.01). The thymus index in the combination group increased significantly (P <0.05). The expression level of CD44 in the combination group was the lowest, which was significantly different from CTX group (P <0.05). Conclusion Fuzheng Xiaohua Decoction can cooperate with chemotherapeutic drugs to increase the sensitivity of tumor cells to the immune system and anti-cancer drugs, and down-regulate or reverse the expression of CD44, a adhesion molecule, in mice with Lewis lung cancer and exert antitumor and anti-metastatic effects.
