目的比较同种异体阔筋膜在不同保存方法下生物学性能的改变,为临床应用提供依据。方法取新鲜同种异体阔筋膜,随机分成三组保存:Ⅰ组梯度乙醇固定保存,Ⅱ组深低温甘油保存,Ⅲ组复合抗生素溶液浅低温保存。于保存后不同时段检测理化特性和生物力学性能,观察其变化,以新鲜阔筋膜作为对照。结果Ⅰ组:保存1个月、3个月时阔筋膜理化特性正常,保存6个月时阔筋膜表面色泽及柔韧性能发生改变,HE染色结果显示,随着保存时间的延长,纤维排列密度逐渐降低。Ⅱ组和Ⅲ组:保存1个月、3个月和6个月时,阔筋膜理化特性均正常。三种方法保存6个月时生物力学性能与新鲜阔筋膜比较无统计学差异。结论三种保存方法均可用于同种异体阔筋膜的保存,在观察期内生物力学性能未见显著性变化。梯度乙醇固定保存在6个月后有形态学变化,深低温甘油保存和复合抗生素溶液浅低温保存则未观察到变化。长时间、大量保存同种异体阔筋膜时不建议使用梯度乙醇固定保存法。
Objective To compare the changes of biological properties of allograft fascia under different preservation methods and provide the basis for clinical application. Methods Fresh allograft fascia was randomly divided into three groups: group Ⅰ preserved with gradient ethanol, group Ⅱ preserved with cryogenic glycerol, and group Ⅲ compound antibiotic solution stored in light temperature. Physical and chemical properties and biomechanical properties were examined at different time points after preservation, and the changes were observed. The fresh fascia was used as a control. Results In group Ⅰ, after 1 month, the physical and chemical properties of the fascia lata were normal at 3 months. The color and flexibility of the fascia lata were changed after 6 months of storage. HE staining showed that the fiber arrangement Decreasing density. Group Ⅱ and Ⅲ: The preservation of 1 month, 3 months and 6 months, the lacrimal fascia physical and chemical properties were normal. There was no significant difference in biomechanical properties between the three methods when preserved for 6 months and fresh fascia lata. Conclusion All three preservation methods can be used for the preservation of allogeneic fascia, and there is no significant change in biomechanical properties during the observation period. Gradient ethanol fixed morphological changes after 6 months, no changes were observed in the preservation of cryogenic glycerol and the preservation of the composite antibiotic solution after mild hypothermia. For a long time, a large amount of preservation of allograft labyrinth is not recommended to use gradient ethanol fixed preservation method.
