该研究的目的在于探索银丹心脑通软胶囊(YDXNT)与运动(游泳)联合作用后对动脉粥样硬化大鼠的预防作用。主要采用二因素(YDXNT,游泳)三水平(0,1,2 g·kg-1YDXNT;0,0.5,1 h游泳)3×3析因设计。结扎大鼠左颈总动脉造成狭窄,然后喂养高脂饲料,建立大鼠动脉粥样硬化模型。8周后,胸主动脉采集血液测定其全血黏度、血浆黏度、纤维蛋白原(FIB)、血清一氧化氮(NO)、血浆6-酮-前列腺素1α(6-keto-PGF1α)、内皮素(ET)、血栓烷素B2(TXB2)水平。取大鼠左颈总动脉组织,进行免疫组织化学染色,观察蛋白SM22α的阳性表达,并对其进行半定量。结果发现,YDXNT结合游泳可以显著降低血浆黏度(F=3.241,P=0.017)、高、低切全血黏度(分别为F=6.444,P=0.001;F=3.002,P=0.024)和FIB(F=4.046,P=0.005)。在升高血清NO,血浆6-keto-PGF1α,降低血浆ET,TXB2方面,交互作用显著(P<0.05)。游泳对于上调蛋白SM22α的表达主效应明显(F=8.088,P=0.001)。该研究提示银丹心脑通软胶囊结合游泳运动后能够改善动脉粥样硬化大鼠的血液流变学指标,具有内皮保护作用,可抑制动脉粥样硬化中的血管重构,对于动脉粥样硬化有积极的预防作用。
The purpose of this study was to explore the preventive effect of YDXNT combined with exercise (swimming) on atherosclerosis in rats. The three factors (YDXNT, swimming) three levels (0,1,2 g · kg-1YDXNT; 0,0.5,1 h swimming) 3 × 3 factorial design. Ligation of the left common carotid artery caused by stenosis, and then fed high-fat diet, the establishment of rat atherosclerosis model. After 8 weeks, the whole blood viscosity, plasma viscosity, fibrinogen (FIB), serum nitric oxide (NO), plasma 6-keto-prostaglandin 1α (6-keto-PGF1α) (ET), thromboxane B2 (TXB2) levels. Left rat common carotid artery tissue was taken for immunohistochemical staining to observe the positive expression of protein SM22α, and its semi-quantitative. The results showed that YDXNT combined with swimming could significantly reduce the plasma viscosity (F = 3.241, P = 0.017), high and low cut whole blood viscosity (F = 6.444, P = 0.001; F = 3.002, F = 4.046, P = 0.005). There was a significant interaction (P <0.05) in increasing serum NO, plasma 6-keto-PGF1α, and decreasing plasma ET, TXB2. Swimming for the upregulation of protein SM22α expression of the main effect was significantly (F = 8.088, P = 0.001). The study suggests that Yin Dan Xin Nao Tong soft capsule combined with swimming can improve the blood rheology of atherosclerotic rats, with endothelial protection, can inhibit vascular remodeling in atherosclerosis, atherosclerosis Sclerotherapy has a positive preventive effect.
