Cotton leaves contain a lot of pigments, which seriously interferes with the isolation and determination of endogenous hormones. This paper reported the use of column and thin layer chromatography (or paper) for purification, followed by identification of auxin, abscisic acid, and cytokinin in cotton leaves by high pressure liquid chromatography and the identification of gibberellin by fluorescence spectrophotometry. Experimental results: 1. The extraction and separation of organic solvent from the adsorption column and the separation process using solvent extraction and column chromatography were basically carried out according to the methods of the literature, except for the phase separation.
