目的采用HPLC法建立复方泽泻胶囊中葛根素和总黄酮的测定方法。方法葛根素测定的色谱条件为Lcromasil C18色谱柱(250 mm×4.6mm,5μm);流动相:甲醇-水(40∶60);检测波长:254 nm;体积流量:1 mL/min;进样量:20μL,柱温:室温。总黄酮采用紫外可见分光光度计在360 nm处测定。结果葛根素在2.032~65μg/mL线性关系良好,平均回收率为97.9%,RSD为1.98%。总黄酮在0~27.50μg/mL线性关系良好,平均回收率为97.1%,RSD为2.98%。结论本方法重现性好,灵敏度高,可用于制剂的质量控制。
OBJECTIVE To establish an HPLC method for the determination of puerarin and total flavonoids in Fufang Zhaoce Capsule. Methods The chromatographic conditions for determination of puerarin were Lcromasil C18 column (250 mm × 4.6 mm, 5 μm), mobile phase of methanol-water (40:60), detection wavelength of 254 nm, volume flow rate of 1 mL / min, Amount: 20 μL, column temperature: room temperature. Total flavonoids were measured using a UV-vis spectrophotometer at 360 nm. Results Puerarin showed a good linearity at 2.032 ~ 65 μg / mL with an average recovery of 97.9% and RSD of 1.98%. The total flavonoids in the 0 ~ 27.50μg / mL good linear relationship, the average recovery was 97.1%, RSD was 2.98%. Conclusion The method has good reproducibility and high sensitivity and can be used for the quality control of the preparation.
