目的利用体外培养重建同基因角膜上皮组织的新方法,为角膜化学伤后重建角膜表面提供理想的角膜上皮移植片。方法以同种或异种脱水保存的浅层角膜基质片作为载体,通过体外培养自体角膜缘上皮细胞,并使其在体外形成复层角膜上皮组织。结果经组织学、细胞超微结构的观察,以及细胞分化蛋白表达的检测,证实体外培养的角膜上皮组织与生理状态下的角膜上皮组织相近。结论体外培养的角膜上皮组织片可作为较理想的角膜上皮移植材料。
OBJECTIVE: To establish a new method of reconstructing corneal epithelium with the same gene in vitro and to provide ideal corneal epithelial graft for the reconstruction of corneal surface after corneal chemical injury. Methods Shallow corneal stroma preserved in the same or different kinds of ascites was used as a carrier to culture corneal limbal epithelial cells in vitro and to form the corneal epithelium in vitro. Results Histology, observation of cell ultrastructure and detection of cell differentiation protein expression confirmed corneal epithelial tissue culture in vitro and corneal epithelial tissue under physiological conditions. Conclusion Corneal epithelial tissue culture in vitro can be used as ideal corneal epithelial graft material.
