目的 探讨凋亡相关基因bcl 2在肺癌中的表达及其与细胞凋亡和增殖的关系。方法 对 3 8例肺癌组织及 2 1例肺部良性组织 ,应用原位杂交技术检测了bcl 2mRNA表达情况 ;应用TUNEL技术对细胞凋亡情况进行了检测 ;应用免疫组织化学法对增殖细胞核抗原 (PCNA)等进行检测。并分别定义凋亡指数和增殖指数。结果 肺癌组织bcl 2基因及PCNA的表达明显高于肺部良性组织 ,P <0 .0 1;肺癌组织中凋亡指数和增殖指数 ( 3 .7%± 2 .4%,13 .9%± 15 .1%)分别高于肺部良性组织 ( 0 .8%±0 .5 %,0 .9%± 3 .2 %) ,P <0 .0 1;bcl 2阳性肺癌组织中凋亡指数 ( 1.9%± 1.2 %)明显低于bcl 2阴性肺癌组织 ( 5 .0 %± 2 .2 %) ,P<0 .0 1;它们的增殖指数均无显著性差异。结论 bcl 2在肺癌中的表达率较高 ,同时在肺癌中细胞凋亡和细胞增殖亦较活跃 ;bcl 2与细胞凋亡存在着密切关系 ,即bcl 2基因过度表达抑制细胞凋亡。bcl 2基因、细胞凋亡和PCNA均可作为肺癌的标志物。
Objective To investigate the expression of apoptosis-related gene bcl 2 in lung cancer and its relationship with apoptosis and proliferation. Methods The expression of bcl 2 mRNA was detected by in situ hybridization in 38 lung cancer tissues and 21 benign pulmonary tissues. The apoptosis was detected by TUNEL technique; Proliferating cell nuclear antigen was detected by immunohistochemistry. PCNA) etc. And define the apoptotic index and proliferation index respectively. Results The expression of bcl 2 and PCNA in lung cancer tissues was significantly higher than that in benign lung tissues, P <0. 01, and the apoptotic index and proliferation index in lung cancer tissues ( 3.7% ± 2.4%, 13.9%). 15.1%) were higher than benign tissue in the lung (0.8% ± 0.5%, 0.9% ± 3.2%), P <0.01; apoptotic index in bcl 2 positive lung cancer tissues. Conclusion The expression of bcl 2 in lung cancer is higher. At the same time, apoptosis and cell proliferation are also more active in lung cancer. bcl 2 is closely related to apoptosis, that is, bcl 2 gene overexpression inhibits apoptosis. The bcl 2 gene, apoptosis, and PCNA can all serve as markers of lung cancer.
