目的:观察大黄■虫丸对球囊损伤内皮诱导的家兔动脉粥样硬化的影响。方法:采用机械损伤动脉内皮合并高脂饲料喂养的方法建立家兔动脉粥样硬化模型。雄性新西兰家兔分为正常对照组,假手术组,模型对照组,阳性对照组,大黄■虫丸低、高剂量组。正常对照组、假手术组与模型对照组给予蒸馏水,阳性对照组给予丹参0.5g/kg,大黄■虫丸低、高剂量组分别给予大黄■虫丸0.9、1.8g/kg。灌胃给药,每天1次,连续60d。常规检查血管璧的组织病理形态和超微结构,采用图像分析系统测定血管壁内膜厚度(IT)和内-中膜厚度(IMT),用分光光度法测定血管壁羟脯氨酸含量。结果:大黄■虫丸能减小动脉粥样硬化斑块面积,减少血管平滑肌细胞增殖和胶原纤维增生,抑制内膜增厚,减轻血管平滑肌细胞线粒体肿胀,减少泡沫样细胞形成;降低IT、IMT值,降低血管壁羟脯氨酸含量。结论:大黄■虫丸有抗动脉粥样硬化作用,机制可能与其疏通经络、活血化瘀、破於生新等功效有关。
OBJECTIVE: To observe the effect of rhubarb insects pill on atherosclerosis in rabbit induced by balloon injury. Methods: Rabbit models of atherosclerosis were established by feeding mechanically injured endothelium and high fat diet. Male New Zealand rabbits were divided into normal control group, sham operation group, model control group, positive control group, rhubarb worm pill low and high dose group. The normal control group, sham operation group and model control group were given distilled water. The positive control group was given 0.5g / kg salvia miltiorrhiza and the rhubarb wormwood low and high dose groups were given Rhubarb insect pearl 0.9,1.8g / kg respectively. Gavage, 1 day, for 60 days. The histopathology and ultrastructure of the vascular wall were routinely examined. The intima - media thickness (IMT) and the intima - media thickness (IMT) were measured by image analysis system. The content of hydroxyproline in the vascular wall was measured by spectrophotometry. Results: rhubarb worm capsule can reduce the area of atherosclerotic plaque, reduce vascular smooth muscle cell proliferation and collagen fiber proliferation, inhibit intimal thickening, reduce mitochondrial swelling of vascular smooth muscle cells and reduce the formation of foamy cells; reduce IT, IMT Value, reduce the content of hydroxyproline in the vascular wall. Conclusion: rhubarb insect tablets have anti-atherosclerotic effect, and its mechanism may be related to its effect of clearing the meridians, promoting blood circulation and removing blood stasis and breaking new ones.
