目的研究转化生长因子-β_1(transforming growth factor beta 1,TGF-β_1)及基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)在重度子痫前期患者胎盘组织的表达及其临床意义。方法 Western blot方法对50例正常妊娠组和60例子痫前期组(25例轻度和35例重度)胎盘组织中TGF-β_1、MMP-9的表达进行分析。其中早发型轻度子痫前期组15例,晚发型轻度子痫前期组10例;早发型重度子痫前期组18例,晚发型重度子痫前期组17例。结果在重度子痫前期组中TGF-β_1的表达明显升高,MMP-9的表达明显降低,与轻度子痫前期组及正常妊娠组相比,差异有统计学意义(P<0.05)。轻度子痫组与正常妊娠组之间比较差异无统计学意义(P>0.05);胎盘组织中TGF-β_1的表达水平和MMP-9的表达水平呈负相关(P<0.05);TGF-β_1、MMP-9在子痫前期患者早发型和晚发型组间的表达两两比较差异均有统计学意义(P<0.05)。结论重度子痫前期患者胎盘组织中TGF-β_1和MMP-9表达异常,提示两者可能共同参与重度子痫前期的发生。
Objective To investigate the expression of transforming growth factor beta 1 (TGF-β 1) and matrix metalloproteinase-9 (MMP-9) in placenta of patients with severe preeclampsia and its clinical significance. Methods The expressions of TGF-β 1 and MMP-9 in 50 normal pregnant women and 60 pregnant women with preeclampsia (25 mild and 35 severe) were analyzed by Western blot. Among them, 15 cases of early-onset mild preeclampsia and 10 cases of late-onset mild preeclampsia; 18 cases of early-onset severe preeclampsia and 17 cases of late-onset severe preeclampsia. Results The expression of TGF-β 1 in severe preeclampsia group was significantly increased, and the expression of MMP-9 was significantly lower than that in mild preeclampsia group and normal pregnancy group (P <0.05). There was no significant difference between mild eclampsia group and normal pregnancy group (P> 0.05). The expression of TGF-β 1 in placental tissue was negatively correlated with the expression of MMP-9 (P <0.05) The expressions of β_1 and MMP-9 in early-onset and late-onset groups were significantly different between the two groups (P <0.05). Conclusion The abnormal expression of TGF-β 1 and MMP-9 in placenta of patients with severe preeclampsia suggests that both may participate in the development of severe preeclampsia.
