为探讨更年春方(GNC)预防实验性原发性骨质疏松的作用机理,采用10~12月龄SD雌性大鼠去卵巢建立骨质疏松模型,用GNC灌药4个月,RT PCR法检测胫骨骨骺ERα、ERβ、OPG、RANKL、osf2 /cbfal与TRAPmRNA的表达。结果:大鼠去卵巢后,血清E2 水平及胫骨骨骺ERα、ERβ、OPG及osf2 /cbfalmRNA的表达均明显下降,RANKL与TRAPmRNA的表达明显增高。GNC治疗后,血清E2 水平无明显变化,胫骨骨骺ERβ、OPG及osf2 /cbfalmRNA的表达均明显增高,RANKL与TRAPmRNA的表达明显下降。提示GNC可能通过上调胫骨骨骺ERβmRNA的表达,有促进成骨细胞、抑制破骨细胞的作用,从而对实验性原发性骨质疏松有预防功效。
To explore the mechanism of Gengnianchunfang (GNC) in preventing experimental primary osteoporosis, 10 to 12 months-old SD female rats were used to establish ovariectomized models of osteoporosis, and GNC was used to infuse the drug for 4 months. RT PCR method was used. The expressions of ERα, ERβ, OPG, RANKL, osf2/cbfal and TRAP mRNA in tibial osteophytes were detected. RESULTS: After ovariectomy in rats, the serum E2 level and the expression of ERα, ERβ, OPG, and osf2 / cbfal mRNA in the tibial bone were significantly decreased, and the expression of RANKL and TRAP mRNA was significantly increased. After treatment with GNC, there was no significant change in serum E2 levels, and the expression of ERβ, OPG and osf2/cbfal mRNA in the tibial osteophyte was significantly increased, and the expression of RANKL and TRAP mRNA was significantly decreased. It is suggested that GNC may improve the osteoblasts and inhibit osteoclasts by up-regulating the expression of ERβmRNA in the tibial osteophyte and thus have preventive effect on experimental primary osteoporosis.
