目的观察大鼠牙移动过程中龈沟液中MMP-1和MMP-9含量的变化。方法在20只6周龄雌性Wistar大鼠上颌一侧安装正畸加力装置,另一侧作为对照侧。每周提取大鼠实验牙及对照牙的龈沟液,并应用SDS-PAGE,Western blot和Gel-Doc凝胶成像系统定量检测龈沟液中基质金属蛋白酶-1(matrix metalloproteinase-1,MMP-1)与基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)含量的改变。采用SPSS11.0统计学软件,对实验各周龈沟液中MMP-1和MMP-9的含量进行方差分析。结果MMP-1和MMP-9含量在实验侧和对照侧均是自加力第1周开始升高(与0周相比P<0.05),第2周时含量大幅增多(与第1周相比P<0.001),至第3周时达高峰,从第4周开始回落,至第7周最低,但仍高于加力前的水平;实验侧MMP-1含量显著高于对照侧(P<0.05);实验侧与对照侧的MMP-9含量差异均无统计学意义(P>0.05)。结论龈沟液中MMP-1和MMP-9的含量在大鼠牙移动过程中先升高后降低,MMP-1较MMP-9与正畸牙移动关系可能更密切。
Objective To observe the changes of MMP-1 and MMP-9 in gingival crevicular fluid during rat tooth movement. Methods Twenty female Wistar rats of 6 weeks old were fitted with orthodontic force device on one side of maxillary and the other side as control. The gingival crevicular fluid was extracted weekly from rat teeth and control teeth. SDS-PAGE, Western blot and Gel-Doc imaging system were used to detect the expression of matrix metalloproteinase-1 (MMP- 1) and matrix metalloproteinase-9 (MMP-9) content changes. SPSS11.0 statistical software was used to analyze the variance of MMP-1 and MMP-9 levels in gingival crevicular fluid during the experiment. Results The levels of MMP-1 and MMP-9 in the experimental and control groups both increased from the first week after the application of force (P <0.05 compared with the 0 week), and increased significantly in the second week (P <0.001), reaching its peak at the third week, falling from the fourth week to the lowest at the seventh week, but still higher than before pre-stress. The content of MMP-1 in the experimental side was significantly higher than that in the control side ). There was no significant difference in MMP-9 content between the experimental side and the control side (P> 0.05). Conclusion The levels of MMP-1 and MMP-9 in gingival crevicular fluid firstly increase and then decrease in rat tooth movement, and MMP-1 may be more closely related to the orthodontic tooth movement than that of MMP-9.
