目的:建立糖皮质激素性骨质疏松症(GIO)大鼠模型,观察不同中药的防治作用。方法:Wistar大鼠132只(雌雄各半),随机分为正常组、模型组、补肾组、健脾组、活血组及骨疏康组。除正常组外,其余各组给予地塞米松(2.5 mg/kg)肌注,每周2次。除正常组及模型组外,其余各组予以相应的中药治疗。造模及给药连续9周。9周后,测定离体股骨骨密度(BMD)及血清骨代谢相关指标。结果:与正常组比较,模型组大鼠BMD下降(P<0.01),血清抗酒石酸酸性磷酸酶(TRAP)的含量升高(P<0.01)。相对于模型组,各治疗组BMD表现出不同程度的升高,其中补肾组升高程度最显著(P<0.01);各治疗组血清TRAP含量显著降低,均有统计学意义。结论:地塞米松诱导的GIO大鼠模型制备成功。补肾中药对GIO具有一定的拮抗作用。
Objective: To establish a rat model of glucocorticoid-induced osteoporosis (GIO) and observe the preventive and therapeutic effects of different traditional Chinese medicines. Methods: 132 Wistar rats (male and female half) were randomly divided into normal group, model group, Bushen group, Jianpi group, Huoxue group and Gushukang group. Except the normal group, the other groups were given intramuscular injection of dexamethasone (2.5 mg / kg) twice a week. In addition to the normal group and model group, the rest of the group to be the corresponding Chinese medicine treatment. Modeling and administration for 9 weeks. After 9 weeks, the indexes of isolated femur bone mineral density (BMD) and serum bone metabolism were measured. Results: Compared with the normal group, the BMD of model group decreased (P <0.01) and the level of serum TRAP increased (P <0.01). Compared with the model group, the BMD of each treatment group showed different degrees of increase, the most significant was the Kidney group (P <0.01). The serum TRAP level of each treatment group was significantly decreased. Conclusion: Dexamethasone induced GIO rat model was successfully prepared. Bushen Chinese medicine on the GIO has a certain antagonistic effect.
