By using an “en bloc” silver-staining method, it was observed under the transmission electron micro-scope that there were various amounts of silver-stained granules of different sizes distributed in the interphase con-densed chromatin. The results analyzed by image analysis instrument indicated that the shape of the larger granules andsome of the smaller granules looked like dots but the others were short fibers. From the results, it could be concludedthat nonhistone protein scaffold existed in the condensed chromatin of root meristematic cell in common wheat, thescaffold proteins were distributed over the whole condensed chromatin in the form of granules and fibers. The relation-ship between chromatin scaffold, chromosome scaffold and nuclear matrix was discussed.
