目的了解白藜芦醇甙(PD)对新生大鼠缺氧缺血性脑损伤(HIBD)大脑皮层脑源性神经营养因子(BD-NF)表达的影响。方法 52只新生7日龄SD大鼠随机分为假手术对照组(NS组)、阴性对照组(NG组)、缺氧缺血组(HI组)、白藜芦醇甙干预组(PD组);通过结扎左颈总动脉及通入8%的氧气2 h制备新生大鼠缺氧缺血性脑损伤模型;各组按照造模后24 h、10 d分为2个亚组,免疫组化法测定各组大鼠大脑皮层BDNF的表达。结果免疫组化结果显示24 h、10 d HI组大鼠皮层BDNF表达(0.144±0.011,0.105±0.011)较NS(0.069±0.005,0.068±0.005)和NG组(0.070±0.003,0.069±0.007)增高(P<0.01),10 d PD组皮层BDNF表达(0.131±0.009)较HI组(0.105±0.011)明显增加(P<0.01)。结论白藜芦醇甙能上调HIBD新生大鼠大脑皮层BDNF的表达,对缺氧缺血后的神经元有保护作用。
Objective To investigate the effects of polydatin on the expression of brain-derived neurotrophic factor (BD-NF) in neonatal rats with hypoxic-ischemic brain damage (HIBD). Methods 52 neonatal 7-day-old SD rats were randomly divided into sham-operation control group (NS group), negative control group (NG group), hypoxic-ischemic group (HI group), resveratrol intervention group ). The model of hypoxic-ischemic brain damage in neonatal rats was established by ligating the left common carotid artery and introducing 8% oxygen for 2 h. The rats in each group were divided into 2 subgroups at 24 and 10 d after operation. Method to determine the expression of BDNF in cerebral cortex of rats in each group. Results Immunohistochemical results showed that the expression of BDNF in cortex of rats in HI group (0.144 ± 0.011,0.105 ± 0.011) at 24 h and 10 d was significantly lower than that of NS (0.069 ± 0.005, 0.068 ± 0.005) and NG group (0.070 ± 0.003, 0.069 ± 0.007) (P <0.01). The expression of BDNF in the cortex of 10d PD group (0.131 ± 0.009) was significantly higher than that of HI group (0.105 ± 0.011) (P <0.01). Conclusion Polydatin can up-regulate the expression of BDNF in the cerebral cortex of neonatal rats with HIBD, and has a protective effect on neurons after hypoxic-ischemic injury.
