目的:探讨组织中糖胺多糖与肿瘤发生的关系。方法:应用自行提纯的硫酸软骨素裂解酶和高效液相色谱(HPLC)二糖分析方法,从细胞外基质代谢的疾病生化学角度,对肺癌患者的病变组织及体液糖胺多糖(GAG)进行了组分和性质的研究。结果:表明肺癌患者组织GAG总量明显高于癌旁的非癌组织;肺癌组织GAG表现为高硫酸化。肺癌患者尿GAG排泄量降低,GAG组分呈严重低硫酸化,GAG的二糖中非硫酸化二糖(△Di0S)含量高于健康对照组15倍。结论:细胞外基质GAG并非仅起单纯的支持和保护作用,还与细胞分化、增殖及其代谢调节等多种生理功能有关。GAG代谢异常与肿瘤的发生、发展过程密切相关
Objective: To investigate the relationship between tissue glycosaminoglycans and tumorigenesis. METHODS: Self-purified chondroitin sulfate lyase and high performance liquid chromatographic (HPLC) disaccharide analysis method were used to analyze the pathological changes of extracellular matrix metabolism and the pathological changes of lung cancer patients and body fluid glycosaminoglycans (GAG). The composition and nature of the study. RESULTS: The total amount of GAG in lung cancer patients was significantly higher than that in non-cancerous tissues adjacent to cancer; GAG in lung cancer tissues showed hypersulfation. The urinary GAG excretion was decreased in patients with lung cancer, and the GAG component was severely hyposulfated. The content of non-sulfated disaccharides (ΔDi0S) in GAG disaccharides was 15 times higher than that in healthy controls. Conclusion: The extracellular matrix GAG is not only a simple support and protective effect, but also related to various physiological functions such as cell differentiation, proliferation, and metabolic regulation.
