目的:对宫颈癌凋亡调控基因Caspase-3进行功能鉴定。方法:采用免疫组织化学技术及RT-PCR方法检测58例宫颈癌术后标本及正常宫颈组织中Caspase-3的表达。结果:与正常宫颈组织相比,宫颈癌组织中Caspase-3表达明显下调,而且Caspase-3表达强度随着病理学分级的升高而减弱,Caspase-3mRNA在正常宫颈及高分化宫颈鳞癌组织中的表达差异无统计学意义(P>0.05),但Caspase-3 mRNA在正常宫颈及中、低分化宫颈鳞癌中的表达差异有统计学意义(P<0.01)。结论:Caspase-3的表达下降可能抑制浸润部位肿瘤细胞的凋亡,增强了肿瘤细胞浸润和转移的能力。
Objective: To identify the function of Caspase-3 in cervical cancer. Methods: Immunohistochemistry and RT-PCR were used to detect the expression of Caspase-3 in 58 specimens of cervical cancer and normal cervix. Results: Compared with normal cervical tissue, Caspase-3 expression was significantly down-regulated in cervical cancer tissues, and the expression of Caspase-3 was decreased with the increase of pathological grade. Caspase-3 mRNA expression in normal cervical tissues and well-differentiated cervical squamous cell carcinoma tissues (P> 0.05). However, there was significant difference in the expression of Caspase-3 mRNA between normal cervix and moderate and poorly differentiated cervical squamous cell carcinoma (P <0.01). Conclusion: The decreased expression of Caspase-3 may inhibit the apoptosis of tumor cells at the infiltrating site and enhance the ability of tumor cells to infiltrate and metastasize.
